چکیده
Abstract Respiratory Syncytial Virus (RSV) is a leading cause of morbidity and mortality in children, due part to their distinct immune system, characterized by impaired induction T-helper 1 (Th1)□immunity. Here we describe cationic adjuvant formulation (CAF)-08, liposomal vaccine tailored induce Th1 immunity early life via synergistic engagement Toll-like Receptor 7/8 the C-type lectin receptor Mincle. Quantitative phosphoproteomics applied human dendritic cells revealed key role for Protein Kinase C-d enhanced cytokine production neonatal identified signaling events resulting antigen cross-presentation. In vivo, single immunization at birth with CAF08-adjuvanted RSV pre-fusion protected newborn mice from infection through antigen-specific CD8+ cells. Overall, novel pediatric characterize its mechanism action providing promising avenue development vaccines against other intracellular pathogens.

Protection against Respiratory Syncytial Virus Infection by DNA Immunization
Respiratory syncytial virus (RSV) remains a major cause of morbidity and mortality in infants and the elderly and is a continuing challenge for vaccine development. A murine T helper cell (Th) type 2 response associates with enhanced lung pathology, which has been observed in past infant trials using formalin-inactivated RSV vaccine. In this study, we have engineered an optimized plasmid DNA ve...
متن کاملInfluenza A virus among the hospitalized young children with acute respiratory infection. Is influenza A co infected with respiratory syncytial virus?
Background: Both influenza A virus (IAV) and respiratory syncytial virus (RSV) cause acute respiratory infection (ARI) in infants and young children. This study was conducted to determine Influenza A virus and its co infection with RSV among the hospitalized children with ARI. Methods: A total of 153 throat samples of the hospitalized young children aged between below one year and 5 years with...

متن کاملImmunization with Hexon Modified Adenoviral Vectors Integrated with gp83 Epitope Provides Protection against Trypanosoma cruzi Infection
BACKGROUND Trypanosoma cruzi is the causative agent of Chagas disease. Chagas disease is an endemic infection that affects over 8 million people throughout Latin America and now has become a global challenge. The current pharmacological treatment of patients is unsuccessful in most cases, highly toxic, and no vaccines are available. The results of inadequate treatment could lead to heart failur...
متن کاملProtection against Helicobacter pylori infection following immunization is IL-12-dependent and mediated by Th1 cells.
The regulatory roles of Th1 and Th2 cells in immune protection against Helicobacter infection are not clearly understood. In this study, we report that a primary H. pylori infection can be established in the absence of IL-12 or IFN-gamma. However, IFN-gamma, but not IL-12, was involved in the development of gastritis because IFN-gamma(-/-) (GKO) mice exhibited significantly less inflammation as...
